Heparan sulfate proteoglycans (HSPGs) are an important class of proteoglycans. They are ubiquitous in the mammalian systems and are involved in a wide variety of biological processes, including regulations of growth factors, cell adhesions and cell-cell communications. Due to the heterogeneity of naturally existing HSPGs, isolation of HSPGs with pure and structurally well-defined glycan structures from natural sources is particularly challenging. To evaluate their biological activities, chemical syntheses of homogeneous heparan sulfate glycopeptides have been developed. However, the overall synthesis is still highly challenging. In order to expedite the synthetic process, herein, we report the establishment of a new chemoenzymatic synthesis strategy of a HSPG mimetic containing the synstatin peptide. Biological functions of the synthetic mimetic will be presented.
