The formation of peptide coverage shell was identified for the first time by adding the amyloid beta 1-40 ( Aβ1-40 ) to the 20 nm and 50 nm gold colloidal particles. Our research team discovered that the two colloidal sizes exhibited completely different features of Aβ1-40 concentration dependence. For the 20 nm gold colloid particle, it had two different induction range at [Aβ1-40 ]/ [Au 20 nm] ≤ 10-5 and [Aβ1-40] / [Au 20 nm] ≤ 10-4 which indicates two step-function responses for gold’s aggregation. However, the 50 nm gold colloid particle had only one induction point at [Aβ1-40] / [Au 20 nm] ≤ 10-4 which indicates one step function. For the 20 nm gold colloid particle, the first step function shows a semi-aggregation of gold and the second step function shows a complete aggregation of gold. In contrast, the 50 nm gold colloid particle gradually aggregates due to the network of Aβ1-40. Based on this observation, our research group is currently proposing the aggregation model which possess a certain threshold coverage.
