Early detection of cancer and vigilant monitoring allow for very favorable treatment outcomes. With the ever-advancing disposal of detection technologies accessible by clinics, it is imperative that arising novel methods be diagnostically accurate, rapid, affordable, and procedurally noninvasive. Circulating non-coding RNAs (ncRNAs) have been attractive biomarkers due to their stability and dysregulated levels during disease. However, extracting and quantifying them are challenging hurdles since ncRNAs can exist in sub-femtomolar concentrations and in complex physiological milieus, where assay sensitivity and selectivity can be compromised. The scope of this work strives to profile and compare microRNA (miRNA) levels from healthy and patient sera in a simple and cost-effective manner. In brief, total nucleic acids are enriched from serum using nano stir bars (nsb’s) and selected miRNAs are quantified via nanomaterial-based exponential amplification reaction (EXPAR). Under optimal conditions, the nsb’s can recover from low fM range and EXPAR responses are resolvable within aM sensitivity. The utilization of nanomaterials to achieve robust and sensitive detection of diseased miRNAs is a promising approach that can broaden the current repertoire of cancer diagnosis.
