Methods for detecting infectious human viruses in environmental samples are critical for protecting human health. This presentation will introduce an Integrated Cell Culture-Mass Spectrometry (ICC-MS) method for non-target screening of infectious viruses in environmental samples. The ICC-MS method was validated and optimized with a mouse coronaviruses spiked in wastewater concentrates. When applied to detect human viruses present in wastewater samples, the ICC-MS method detected mammalian reoviruses in influent, wastewater treated with secondary treatment, and final effluent after UV disinfection. Strain-specific viral peptides from reovirus type 1, type 2, and type 3 were consistently detected in primary influent and wastewater treated with secondary treatment. Infective reviruses were also detected in final effluent post-UV treatment. Our work has demonstrated that the ICC-MS method is a promising technique for monitoring infectious human viruses in environmental samples without the need for primer design.
