Recent studies have presented the new ionization technique termed vibrating sharp-edge spray ionization (VSSI). In its simplest form, VSSI consists of a vibrating substrate that nebulizes a bulk liquid sample. In one embodiment, a VSSI source is comprised of a fused-silica capillary that is attached to the vibrating substrate (cVSSI). Experiments have shown the approach to be highly robust for generating small droplets (&lt;10 μm) across a flow range spanning &gt;3 decades making VSSI highly suitable for coupling with liquid chromatography separations.  Recently, cVSSI has been demonstrated as a new tool for performing in-droplet hydrogen / deuterium exchange (HDX) reactions.  This methodology is demonstrated as a new means to identify compounds in complex mixtures.  It is proposed that this be accomplished using a database matching approach in which isotopic distributions for compounds that have undergone in-droplet HDX are examined.  Additionally, the new approach is highly suited for the prediction of isotopic distributions for compounds that may not be present in any database.  The advantages and limitations of cVSSI HDX will be discussed.
