Quantum dots (QDs) have been highlighted in biomedical applications due to its narrow emission band, long-term photostability, and high brightness. However, its bioapplications are limited because it is made up of heavy metals such as cadmium and lead which is highly toxic. To compensate the defect, ZAIS[(ZnxAgyInz)S2] nanoparticles (NCs) synthesis was developed using the ultrasonic method. ZAIS can control wavelength by changing composition and also have low toxicity since they do not contain heavy metals. Following ligand exchange processes from oleylamine (OA) to mercaptopropionic acid (MPA), the synthesized NCs were coated with Bovine Serum Albumin (BSA) for hydrophilicity and biostability. MPA-coated ZAIS (ZAIS-MPA) and BSA are expected to conjugate through a strong physisorption with several forces. We observed the tendency in electrophoresis by adding NaCl (electrostatic attraction), urea (hydrophobic interaction), and sodium dodecyl sulfate (SDS) to the ZAIS-MPA-BSA to determine the bond origin between ZAIS-MPA and BSA, as well as to determine which forces were dominant. We also observed changes in albumin concentrations vs. ZAIS through electrophoresis to find the optimal concentration of antibodies for bioapplications. Finally, we added Herceptin to NCs (ZAIS-MPA-BSA) for the target specific imaging of breast cancer cell (SK-BR-3) which overexpresses Her-2 receptors. Her-2 receptors of SK-BR-3 cells were imaged to evaluate its affinity to Herceptin conjugated ZAIS NC, and its distributions as well as dynamic behaviors of HER-2 receptors on SK-BR-3 cell surfaces.
