As an efforts to understand the effects of surface topography and chemistry on the foreign body reaction against PDMS implants, PAA micropatterns were grafted onto PDMS surface via surface-initiated photopolymerization. As model micropatterns, 100 μm-wide and 3 μm-thick hole and stripe micropatternswere grafted onto PDMS surface. Grafiting of PAA micropatterns createdtopographical cues by hole or stripe micropatternsas well as chemical cues by hydrophilic PAA microdomains. According to in vitro studies, PAA-grafted PDMS surface had lower proliferation of both macrophages and fibroblasts regardless of pattern presence. However, PDMS with PAA micropatterns, especially stripe micropatterns, inhibited most proliferation of fibroblasts and differentiation into myofibroblasts. In vivo studies have shown that PDMS samples with stripe micropatterns differentiate macrophages into inflammatory M2 macrophages. In addition, stripe micropatterns showed the best results in terms of inflammation score, TGF-b expression, number of macrophages and myofibroblasts, and collagen thickness and density.
