Breast cancer is the most common cancer diagnosed among women in the United States. Due to the high rates of progression and recurrence, there is an urgent need for novel therapeutic approach in the treatment of breast cancer. Sonodynamic therapy (SDT) has emerged as a promising, non-invasive treatment for cancerous tumors. SDT utilized the combination of a sensitizer, ultrasound and molecular oxygen. Herein, we report a nanoparticle-based sensitizer that can be activated using ultrasound to generate reactive oxygen species. Silver-embedded mesoporous silica nanoparticles loaded with sensitizing molecules display a significant decrease of the breast cancer cell proliferation when compared to free sensitizing molecules under ultrasound irradiation. Moreover, 1,3-diphenylisobenzofuran (DPBF) and terephthalic acid (TA) were used to monitor the generation of the reactive oxygen species. This study demonstrates that these novel therapeutic agents greatly improve the effectiveness of the SDT treatment with minimal side effects.
