Lengthy cycle times in discovery of drug candidates, compounded by high levels of attrition as they progress through clinical development, severely hampers efficiency in Pharmaceutical R&D.  Excellence in synthetic chemistry has the potential to be a key mitigating factor capable of increasing probability of success and accelerating programs through all stages of their life cycles, particularly in discovery and early development where synthesis is often on the critical path.  In response to this guiding principle, we have established Discovery teams integrating traditional medicinal and process chemistry mindsets, as a way to augment synthetic chemistry capabilities and expertise.  Through this architecture, early identification and resolution of chemistry bottlenecks improves speed of execution of SAR, breadth of chemical space explored, and rapidity in transition to development of high quality candidates.  Relevant examples in support of the Merck pipeline via this approach will be discussed.<br/>
