Nanopore sequencing, which directly reads sequences from individual DNA or RNA strands, is routinely carried out with enzymatic ratcheting strategies. Nanopore sequencing is superior to other platforms by providing a long read-length along with rich information of epigenetic modifications and base lesions directly during sequencing. However, the utilization of motor enzymes during nanopore sequencing has restricted its uses in sequencing DNA or RNA at this moment. Further development of nanopore sequencing for other biomolecules such as xeno nucleic acid (XNA), poly-peptide or poly-saccharide requires laborious effort in screening compatible motor enzymes required for sequencing. Nanopore Induced Phase-Shift Sequencing (NIPSS), which is a variant form of nanopore sequencing, could sequence any biomolecules but with a short read-length. As a proof of concept, we report direct sequencing of a sugar-modified XNA, which can’t be directly sequenced by any existing platforms but urgently required for applications like gene therapies and aptamer screenings. The demonstrated results has approved the feasibility of NIPSS, which suggests its uses in sequencing a wider variety of biomolecules.
