A logically derived sequence (LODES), based on the understanding of the dissociation mechanism of monosaccharides and disaccharides, was developed for tandem mass spectrometry to determinate the structures of underivatized oligosaccharides. Diastereomers of each monosaccharide, anomericity, and linkage position of each glycosidic bond can be determined using this new method. This method is rapid and has high sensitivity. It can be applied for in situ structural determination of oligosaccharides eluted from HPLC. We have applied this new method to determine the free oligosaccharides in bovine milk. More than 20 free oligosaccharides in bovine milk which have not been reported before were identified. The biosynthesis pathways of these newly discovered oligosaccharides will be discussed.
