A well designed nanopore sensor could resolve a variety of information from single molecules such as small molecules, nucleic acids or proteins. The acquired event information, which results from different conformations, charged states, chemical bond formation or molecular vibrations of the analyte, forms the basic criteria for molecular identifications. However, it is a technical challenge to achieve a fabrication resolution with atomic precision when making a solid state nanopore. Alternatively, the building components of biological nanopores could be well-manipulated via site directed mutagenesis but restricted to a limited combination of 20 natural amino acids. Here we report a novel concept of nanopore fabrication with designed bio-inorganic interfaces by taking a biological nanopore as an engineering template. The designed bio-inorganic interfaces, which provide flexible tuning of the nanocavity geometry, introducing specific pore-analyte interactions or switching the charged state of a nanopore restriction, have significantly broadened the field of applications using nanopores for direct single molecule sensing.
