Functionalized lipids are useful tools to interrogate lipid function in biological processes. We have recently added photoswitchable lipids to this toolset, which allow for the control of lipid function with the spatiotemporal resolution of light. In two recent studies, we demonstrated optical control of TRPV1 channels through photoswitchable fatty acids, termed FAAzos and optical control of PKC (and Munc13) through photoswitchable diacylglycerols, termed PhoDAGs. Herein, we present the expansion of this approach to a series of lysophospholipids, including sphingosine and sphingosine-1-phosphate (S1P). We present efficient synthetic routes toward the respective photoswitchable analogs and demonstrate reversible control of molecular targets both in the signaling and the metabolism of these lipids in mammalian cells and in mice.<br/>
