N1-cyclic adenosine diphosphate ribose (cADPR) and N1-cyclic inosine diphosphate ribose (cIDPR) are calcium-releasing second messengers, yet despite decades of study the mechanism of action of cADPR, cIDPR, and their analogs remains poorly understood. Our earlier studies using variable-temperature 1H NMR provided a detailed picture of the conformational landscape of cADPR and two analogs; in this talk we will present our recent results on the synthesis and conformational analysis of several known and new cIDPR and cADPR analogs. Additionally, our progress on the synthesis and NMR characterization of a potential photoaffinity label to be used for cADPR receptor identification studies will be presented.
