Epidemiological studies have linked exposure to nanoscale particulates from ambient and occupational sources with increased morbidity and susceptibility to lung infections, including pneumonia.  Advances in quantitative global proteomics and redox proteomics on thiol-based posttranslational modifications now make it feasible to investigate cellular pathways and even the precise sites of cellular protein redox modifications in response to nanomaterial exposure, and link these to modulation of macrophage function.  In this presentation, we will discuss our recent work applying advanced quantitative redox proteomics and global proteomics tools to identify cellular pathways and molecular targets of redox stress caused by different types of metal oxide nanoparticles. Both adaptive and stress-specific pathways have been identified.  The resulting data on early molecular initiating events with engineered particles not only have the potential to advance predictive hazard capability for helping to guide safe nanotechnology, but also provide an important mechanistic understanding of susceptibility associated with nanoparticle exposure.
