Bulky a,β-dehydroamino acids (bulky △AAs) are proven to increase the proteolytic stability of b-hairpins when inserted into the turn regions of these peptides. However, their impact on the stability of helical secondary structures is less explored. We hypothesized that bulky DAAs could protect 310-helical peptides from proteolysis due to the similarities between 310-helices and b-turns. Our efforts to evaluate this hypothesis by inserting bulky DAAs such as dehydrovaline into simple known 310-helical peptides will be described. We ultimately envision utilizing this method to increase the proteolytic stability of therapeutic peptides.
