Depression as one of the most common and disabling mental disorders, with a worldwide prevalence of approximately 17%. However, understanding of the pathophysiology of depression is still rudimentary due to its complex aetiology. Therefore, it is of great significance for revealing the mechanism of depression by accurately detecting the bioactive molecules, and uncovering their regulatory effects. Two-photon (TP) fluorescence imaging is one of the most appropriate tools for brain imaging. Thus, we construct a series of new fluorescent probes with high sensitivity and selectivity, and realized the in situ two-photon fluorescence imaging of depression-related bioactive molecules, such as acetylcholinesterase (J. Am. Chem. Soc. 2019, 141, 2061−2068), hydroxyl radical (Angew. Chem. Int. Ed.2019, 58, 4674–4678), ozone (Chem. Sci.2019,10, 2805-2810), biothiols (Analyst, 2019, 144, 191-196.) and so on. Utilizing these probes, we systematically investigated the changes of those active molecules and explored the signal pathway related to depression, which provide important information and ideal tools for expanding the understanding of pathogenesis of depression.
