In the present study, we have designed and synthesized a series of new nitroxide derivatives, and examined their anti-inflammatory and analgesic activities. Considering the beneficial effects of low-level laser irradiation (LLLI) therapy, we proposed that a combination therapy of low-level laser irradiation with new nitroxide derivatives might lead to an effective therapeutic strategy for the treatment of acute limb ischemia/reperfusion injury. Herein, the effects of the combined treatment on limb I/R induced tissue damage were evaluated by the measurement of biomedical parameters: MDA, MPO and TNF-alpha levels. In addition, we also investigated the effects of the combined treatment on remote organ be monitoring the serum AST, ALT, BUN, and Cr levels. Interestingly, when the LLLI was employed in combination with an analgesic anti-inflammatory agent (5j), the protective effects of the combined treatment against I/R injury were significant.<br/>The combined treatment was found to significantly attenuate the structural and functional damage observed in limb I/R. This observation is of clinical interest because it demonstrates that the combination of LLLI and an analgesic anti-inflammatory agent may provide an effective<br/>therapeutic strategy against reperfusion damage.
