In order to investigate the effect of purity and heterogeneity of a peptide sequence used to direct nanoparticle (NP) synthesis, palladium nanoparticles on the order of 2 nm were synthesized. The soluble fraction of bacterial lysates containing repeat units of a metal binding domain (Pd4) fused to the carrier protein Green Fluorescent Protein (GFPuv) was used to direct nanoparticle synthesis. Our data show that the synthesized NPs can be used for cross-coupling reactions such as Suzuki and Stille cross-coupling. Due to their high stability and the capability of using green solvents, impure recombinant peptide-directed Pd NPs may serve as a remarkable catalyst for coupling reactions. Characterization of palladium NPs confirmed the synthesis of highly ordered materials, as evident by microscopy and elemental analysis.
