Abstract To produce new antimicrobial agents with better antimicrobial profile and to explore the potential of quinolone imine derivatives as anti Gram positive and Gram negative agents, we synthesized some novel sparfloxacin analogues by introducing imine functionality at C-4 position. Molecules having an azomethine group are known as imines (Schiff bases). The chelating abilities of these compounds can be utilized to attach the antibiotic to a specific site from which it can interfere with the bacterial proliferation. In view of that, we prepared few imines through reductive amination of ring ketone at C-4 position of sparfloxacin with amine group of amino acetic acid and para-bromo aniline (Compound A and B respectively). To synthesize compound C, primary amine group of sparfloxacin was reacted with salicylic aldehyde. The structure of these derivatives was established by various analytical techniques i.e., UV, IR, and mass spectrometry. Among the synthesized compounds, A and B showed decreased activity as compared to sparfloxacin. Compound C exhibited better activity than sparfloxacin against Gram –ve P. aeruginosa while decrease in activity was observed against B. subtilis and S.aureus. New derivatives showing activity better than the parent Sparfloxacin can be used to eradicate resistant microbes.
