Gluathione (GSH) plays important role in several biological systems such as being antioxidants in defense against free radicals and toxins, involved in modulation of cell proliferation, maintaining the proper thiol-disulfide status of proteins, etc. The abnormal level of GSH in humans is responsible for various diseases. Therefore, there is great need to develop a sensor for the selective and sensitive detection of GSH. A fluorescent sensing platform has been developed for the detection of GSH on the basis of carbon dots (Cdots) and MnO2 nanospheres of different sizes. Highly fluorescent Cdots were synthesized from ascorbic acid by using microwave method. MnO2 nanospheres of different sizes were prepared from different concentration of methionine and fixed concentration of KMnO4. The size and shape of Cdots and MnO2 nanospheres was confirmed by HRTEM analysis. MnO2 nanospheres of different sizes in the presence of Cdots leads to quench the fluorescence intensity, quantum yields, and average lifetime values due to occurrence of fluorescence resonance energy transfer (FRET) mechanism between the Cdots and MnO2 nanospheres. Cdots act as electron donor and MnO2 nanospheres act as electron acceptor in this prepared sensing platform. The detection of GSH has been performed by using Cdots with different size MnO2 nanospheres as sensing platforms. The redox reaction occurs between MnO2 nanospheres and GSH, which result in conversion of MnO2 to Mn2+. The variation in rate of quenching (“turn-off”) and recovery (“turn-on”) of fluorescence intensity of Cdots is dependent on the different size of MnO2 nanospheres. MnO2 nanospheres of large size showed best detection limit of 15 µM for the detection of GSH. The prepared sensing platform is fast, economical, and environment friendly for the detection of GSH.
