Natural bio-polysaccharide-based hydrogels combined with magnetic materials shows significant advantages in biomaterials for medicine and in vivo applications due to its biosafety, remote-control ability and remote heating ability. However, the use of unsafe crosslinking agent and reagents and complex operation steps and instability limited its application in the field of human therapy. In this study, we used a one-step method to prepare a partially deacetylated chitin nanofibers (DEChNs) based magnetic hydrogels by ammonia gas phase gelation. The formation of the gel is formed by the network of DEChNs nanofibers crosslinked by hydrogen bonds. The Fe<sub>3</sub>O<sub>4</sub> magnetic nanoparticles (MNPs) with a dimeter of ~10 nm were deposited on the surface of DEChNs through coprecipitation. The basic properties of the prepared magnetic hydrogels were characterized by XRD, FTIR, XPS, SEM, TEM, VSM. The mechanical property of the magnetic hydrogels were tested by rheometer and the storage modulus (G’) reached up to 5500 Pa. The prepared magnetic hydrogel showed remolding ability by adjusting the pH value assisted with ultrasound treatment and ammonia gas phase gelation. The biocompatibility of magnetic hydrogels was evaluated by ATDC-5 mouse chondrocytes and the cells can adhesion and proliferation on the surface of the magnetic hydrogel. In addition, the magnetic hydrogels showed heating behavior in alternating electromagnetic fields (AMFs) and the temperature can be adjusted by changing the content of magnetic particles inside. HCT116 colon cancer cell can be killed in-vitro when subjected to M-DEChN hydrogel under AMF treatment, which shows application prospects in adjuvant therapy for cancer.
