Background: Currently, there are no effective therapeutic drugs available to protect or limit the intestinal epithelial damage observed in inflammatory bowel disease (IBD). Several clinical and preclinical studies have shown probiotics as an emerging novel therapeutic strategy for IBD; however their probable mechanisms are still poorly understood. Although Lactobacillus acidophilus (LA) improves disease severity associated with IBD, a very little is known about the specific role of cell wall components of LA in IBD. Herein, we investigated the protective effects of cell wall components Lactobacillus acidophilus in lipopolysaccharide (LPS)-induced murine colitis. Methods: LPS-sensitized rats were modeled colitic by colonic administration of LPS (500 µg/rat) for 14 consecutive days. Concurrently, cell wall components isolated from 103, 106 and 109 CFU of LA was injected subcutaneously for 21 days, considering sulfasalazine (100 mg/kg, p.o.) as standard. The severity of colitis was assessed by body weight loss, food intake, stool consistency, rectal bleeding, colon weight/length, spleen weight and histological analysis. Colonic inflammatory markers such as myeloperoxidase activity, C-reactive protein and pro-inflammatory cytokines while oxidative stress markers viz. malondialdehyde, reduced glutathione and nitric oxide were also assayed. Results: Cell wall components isolated from 106 and 109 CFU of LA significantly ameliorated the severity of colitis by reducing body weight loss and diarrhea & bleeding incidence, improving food intake, colon weight/length, spleen weight and microscopic damage to the colonic mucosa. The treatment also reduced levels of inflammatory and oxidative stress markers and boosted anti-oxidant molecule. However, cell wall components isolated from 103 CFU of LA was ineffective in modulating disease severity. Conclusion: Cell wall components isolated from 109 CFU of LA produced more prominent effect to attenuate LPS-induced colitis by limiting immuno-inflammation and oxidative stress.
