It is of great importance to applied efficient signal amplification strategy to nucleic acid-based electrochemiluminescence (ECL) biosensor for more excellent analytical performance. In this work, an excellent signal amplification strategy was suggested by link the aptazyme-mediated enzyme-amplified reaction to the catalyzed hairpin assembly (CHA)-mediated isothermal enzyme-free exponential signal amplification systems. The verification result of biological and ECL methods shows that the cascaded signal amplification systems could successfully converted and amplified the amount of adenosine triphosphate (ATP) to duplex DNA. The suggested strategy was successfully combined with quenching effect of ferrocene to fabricated a novel sensitive “signal-off” ECL biosensor for quantitative detection of ATP. At the optimized conditions, the proposed “signal-off” ECL biosensor exhibited good stability and specificity for ATP detection in the range from 0.1-1000 pM with a lower detection limit of 0.054 pM (S/N=3). Moreover, this biosensor was also demonstrated satisfactory recovery from human serum samples, and successfully applied to the intracellular ATP detection of <i>Staphylococcus aureus</i>. In a word, this work provides a promising method for ATP detection and arousing the interest to applied such strategy to improving ECL sensing performances.
