Vaccination with bacterial surface polysaccharides and their protein conjugates has been highly effective in providing protection to human against numerous infectious diseases. The current practice of producing bacterial polysaccharide-based vaccines by extraction and purification of heterogeneous polysaccharides from bacterial cultures can be improved to overcome challenges in quality control and undesired complications in application. One strategy is to synthesize structurally defined oligosaccharides and analogs that may have enhanced efficiency in addition to minimizing quality control complications and reducing side facts. We have developed efficient chemoenzymatic methods for synthesizing structurally defined oligosaccharides and analogs of bacterial surface polysaccharides which can lead to improved vaccine candidates. The structurally defined probes are also essential for better understanding the important roles of these biomolecules. I will present our recent efforts on developing chemoenzymatic synthetic methods for synthesizing oligosaccharides and analogs of several bacterial surface polysaccharides and their potential applications in probing protein recognition and vaccine development against bacterial infection.
