Herein, we will report the development of an acid cleavable phosphoramidate linker for controlled-release of payloads for antibody- and small molecule-drug conjugates applications. This water-soluble linker was found to have tremendous stability at physiological pHs while rapidly releasing its payload at acidic pH. We have also tailored the linker to release payloads of diverse functional groups (from cytotoxic compounds to turn-on and ratiometric dyes), broadening its applications.
