Common treatments for malaria caused by Plasmodium falciparum have been found to lack efficacy when drug-resistant strains or certain pre-existing health conditions are present. Because current treatments for diseases caused by Plasmodium falciparum are often inadequate, the development of new treatments is important to the welfare of those exposed to these parasites. Pyrimidines are heterocyclic, sharing this trait with many drugs used for malaria treatment. With their resemblance to current drugs certain pyrimidine derivatives may show activity against malaria. 3,4- and 3,5-difluorophenylpyrimidines were synthesized via the reaction of 3,4- and 3,5-difluorophenylvinamidinium salts with select amidines and tested against three Plasmodium falciparum strains, known to have resistance to common antimalarials. All compounds were found to be nontoxic by TX2 in vitro assay, but most showed low degrees of potency against the selected strains. The complete results for all compounds will be presented.
