Integrins play a key role in facilitating immune cell trafficking throughout the body, and provide compelling opportunities to impact serious diseases. A breakdown in the trafficking of circulating leukocytes to the bowel contributes to the development of both ulcerative colitis (UC) and Crohn’s disease (CD). The α4β7 integrin is a clinically validated target for these diseases, with the humanized α4β7 monoclonal antibody vedolizumab (Entyvio) providing an important life-changing therapy for IBD patients. A series of oral small molecule α4β7 antagonists have been identified and evaluated. The data and details from this program will be presented.
