Cancer therapeutics often compromise the immune system of patients, making microbial infections one of the most common complications during treatment. Thus, researchers are prompted to develop medicines that serve a dual purpose; combating both cancer and opportunistic infections. Medications containing the imidazole moiety, like clotrimazole (anitfungal) and dacarbazine (anti-cancer), are currently being used in a clinical setting as both anti-tumor and antimicrobial agents. But researchers attempting to unify both types of anti-proliferative activities have found most imidazole derivatives to be relatively insoluble in aqueous media. The triazole heterocycle differs by one nitrogen, and potentially has a greater dipole moment in comparison to imidazole. In addition, forming a salt complex further increases the hydrophilicity of the organic compound; making the triazole heterocycle an excellent foundation for research and drug development. This multidisciplinary project will involve the synthesis and characterization of N,N’-bis-substituted 1,2,3-benzotriazolium salts to further elucidate the structure activity relationship in ESKAPE (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter) pathogens. A series of lipophilic and hydrophilic substitutes will be substituted on the parent molecule 1H-1,2,3-benzotriazole to determine the optimum salt that plays a role in the cessation of ESKAPE bacterial species and cancer cell lines.
