Speciation is an excellent tool that provides a detailed analysis of chemical transformations of metal-based therapeutics to better understand their mode of action. This presentation covers the importance of speciation in three classes of metal-based therapeutics, two classes of coordination complexes and polyoxometallates, under conditions of their respective biological studies. The first vignette focuses on the importance of the speciation chemistry of monosubstituted decavanadates to evaluate their inhibitory effects against Mycobacterium smegmatis. Decavanadate ([V10O28]6-, V10) has been found to be a more potent inhibitor of Mycobacterium smegmatis than monovanadate (VO43-, V1), and, as such, the growth inhibitory effects of substituted decavanadates are of interest. Therefore, the inhibitory effects of two monosubstituted decavanadates, such as monomolybdo(VI)nonavanadate(V) ([V9MoVIO28]5-,2 V9Mo) and monoplatino(IV)nonavanadate(V) ([H2PtIVV9O28]5-, V9Pt),3 were investigated and evaluated based on the 51V NMR speciation data. Time-dependent 51V NMR speciation studies confirmed that V10 is the most potent inhibitor, closely followed by V9Pt due to their exceptional stability in the 7H9 growth medium of M. smeg. The second vignette focuses on the importance of time-dependent studies of vanadium(V) dipicolinates, the first class of coordination complexes to enhance the oncolytic viruses for immunotherapy applications. The third vignette covers the synthesis, characterization, and time-dependent speciation of the novel antidiabetic V(IV) diamine complex. Speciation and stability of the complex were investigated by carrying out 51V NMR, EPR, and UV-Vis studies. All vignettes demonstrate how speciation demystifies the complexity of the biological vanadium chemistry.
