Increasing regulatory pressure and the development of resistant fungal pathogens to market-leading fungicides has created significant need for new crop disease solutions in the global fungicide market.  Florylpicoxamid is a novel, broad-spectrum fungicide developed by Corteva Agriscience to address these challenges. The discovery effort that led to this new fungicide was inspired by the natural product UK-2A, a macrocyclic picolinamide. Incorporating starting materials from the natural pool of chiral molecules, a modular approach to synthesizing 9- and 10-membered lactone analogs of UK-2A was undertaken and an understanding of key structure activity relationships (SAR) developed. Leveraging this SAR in the design of simpler, more accessible acyclic analogs ultimately enabled the discovery and development of Florylpicoxamid.
