The flow-batch technique Lab-In-Syringe is the latest offspring of the flow automation approaches. It is based on using the void of an automated syringe pump as a size-adaptable yet steadily sealed mixing, reaction, and extraction chamber. It allows straightforward automation of laboratory procedures, above all those directed to analyte extraction and matrix elimination.<br/>High procedural versatility of this tool is yielded by placing a magnetic stirring bar inside the syringe and software-aided operation control. This enables homogenous and on-demand in-syringe mixing of step-by-step added reagents as needed widely independent from solution volumes and viscosities. It also enables to bring immiscible phases into thorough contact allowing the automation of dispersive liquid-liquid extractions. So far, LIS has been used for the automation of practically all standard procedures including single-drop microextractions, dispersive solid phase extraction, cloud point extraction, or head-space separations that were surveyed in a recent tutorial to this technique.<br/>We give an overview on three methodologies and operation modes how this technique can be used for the automation of sample pretreatment protocols coupled online to liquid and gas chromatography. The automated extraction and separation protocols were applied in the determination of aromatic contaminants, sulphonamide antibiotics, and nonsteroidal anti-inflammatory drugs.
