Applications of liquid chromatography and liquid chromatography-mass spectrometry in the biosciences span a huge range of analyte sizes and polarities. At the low molecular weight end of the spectrum are individual amino acids, sugars, and nucleobases, and at the upper end are various types of proteins, including monoclonal antibodies and their constructs. While C18 columns remain popular for many peptide and protein separations, they often lack sufficient retention or selectivity for applications outside this realm. This presentation will outline several novel chromatographic applications used in development and characterization of reference materials at NIST, with a particular focus on standards for biotherapeutics and clinical diagnostics.
