There is an urgent need for new antifungal drugs. The enzyme Acetyl CoA Synthetase 1 (ACS1) is essential to fungi but not to human which makes it an excellent antifungal drug target. Substrate analogs, AMP alkyl esters were synthesized, screened against ACS1 enzymes from four different fungal organisms. Several substrate analogs exhibited inhibition of ACS1 at nanomolar concentration.
