Nucleotide phosphorothioates have been identified as potential anticancer, antiviral and antimicrobial agents, but their synthesis has been proven challenging. The syntheses of  2'F-adenosine-5'-monothiophosphate and 3'F-guanosine-5'-monothiophosphate will be described. The talk will feature several organocatalyzed and biocatalyzed reactions including two novel methods for the diastereoselective installation of fluorine,  new catalysts for a key silyl transfer reaction and the thiophosphorylation of the guanosine nucleoside, as well as a highly efficient and diastereolective reduction of a fluoroketone by a KRED enzyme.
