The sustained success of vector control requires the continued delivery of new classes of chemical insecticides compatible with a wide variety of potential end uses, including long-lasting nets, indoor and outdoor sprays, toxic baits, and larvicide technologies. Globally, vector control relies on several molecular modes of action and classes of insecticides recognized by the Insecticide Resistance Action Committee (IRAC). Natural products (NPs) and their derivatives represent an important source of new insect-active chemistries with potential to meet the Target Product Profiles (TPPs) required of new vector control products. This presentation will review an image-based phenotypic screening paradigm employing automated digitization and wide exploration of the NP chemical space to identify new chemistries for vector control. The diversity and utility of phenotypic endpoints for discovery of new Active Ingredients (AIs) with contact and spatial efficacy against mosquitoes will be discussed. The recovery of chemistries that induce sub-lethal, delayed-effects, and implications for control will be explored. Discoveries will be reviewed in the context of the opportunities and challenges associated with the NP product registration pathway.
