Biomaterials are increasingly used in tissue engineering for their desired properties such as, biocompatibility, biodegradability, porosity and injectability. Low-acyl gellan gum (LA-GAGR) and hyaluronic acid (HA) were ionically crosslinked by tricalcium phosphate (TCP), and an injectable gel composite that slowly decomposes over time was fabricated. Additionally, the diffusion of LA-GAGR and mini-GAGR (a fragment oligomer of gellan gum) through bone was studied to obtain diffusion coefficients. To investigate how LA-GAGR and mini-GAGR interact with bone marrow stem cells after diffusion, the osteodifferentiation signalling pathway of bone marrow stem cells was investigated, as well as collagen type I production. Moreover, antibacterial tests showed that the minimum inhibition concentration of LA-GAGR is 3% against gram-negative bacteria Pseudomonas aeruginosa using the Kirby Bauer method. However, when TCP is added at 0.5%, LA-GAGR showed bacterial inhibition at 1% concentration. LA-GAGR shows great potential as an injectable low-cost biomaterial for bone regeneration strategies.
