Glyphosate is the most widely used non-selective and broad-spectrum herbicide in the world. Classification of glyphosate as a carcinogenic agent by international and regional regulatory bodies have garnered interest in its fate and transport in groundwater connectivity. However, the fate and trace analysis of glyphosate and its metabolites is cumbersome due to its polar and zwitterionic properties. We investigated the transport and interaction of glyphosate, glufosinate and AMPA in karst watersheds. We developed and utilized a vigorous LC-MS/MS method for the quantitation of glyphosate and its metabolites in environmental waters of through reversed-phase and chromatographic separation technology on an anion-exchange column.  Various agricultural surface and ground water samples from Central Kentucky were used to demonstrate and validate the process. Results indicate an increase in sensitivity and stability for analysis of derivatized glyphosate on an LC-MS/MS.
