We present a continuous operation of open circuit potential (OCP) measurement based glucose sensor, employing direct electron transfer type FAD dependent glucose dehydrogenase (FADGDH). The glucose concentration was successfully determined based OCP signal for 3 months, under continuous operation.<br/>To control glycemic levels of patients with type 1 diabetes, continuous monitoring of blood glucose levels is inevitable for the subsequent infusion of insulin. Current commercially available CGM sensors employing amperometric enzyme sensor principles, which are operated under continuous potential application to the glucose oxidoreductases and oxygen or redox mediator as the electron acceptors, with continuous turn-over of the catalytic reaction. Under such harsh conditions, inactivation of enzymes and mediators are unavoidable during the continuous operation. Additionally, the potential application to the system may easily cause the oxidation of interfering substances directly on electrode surface. To overcome these limitations of current technologies, we have reported OCP based enzyme glucose sensing principle. The OCP measurement principle greatly improved sensor stability by minimizing enzyme inactivation with OCP sensing principle without being affected by interferences such as ascorbic acid and acetaminophen. In this study, we demonstrated a long-term operation of OCP based glucose sensor for real-time monitoring through continuous operation for 3 months. OCP signal increased depending on glucose concentration (0.1 mM~20 mM) and the sensor was operated continuously without showing any significant decrease on the slope of calibration curve for 108 days. Since the longest life time of enzyme based commercially available CGM sensor is only 14 days, this result indicated that the OCP principle can allow to develop continuous glucose monitoring enzyme sensor which can provide super stable and accurate sensor signal for long-term period.
