Our laboratory has studied the biosynthetic pathways of trans-acyltransferase polyketides from Bacillus subtilis and Bacillus amyloliquefaciens including bacillaene. The biosynthesis of bacillaene has several unusual features, including β-branching, a trans-enoylreductase, and a cytochrome P450 enzyme. These unique biosynthetic features, plus the instability of bacillaene, have made it difficult to characterize and understand the biosynthetic pathway of this polyketide. The polyketide synthase product from this pathway is dihydrobacillaene, which lacks a certain double bond on bacillaene, and it has been expected that this is oxidized by an unknown pathway to produce bacillaene. Previous research from our lab has shown that the cytochrome P450 enzyme, PksS, will react with dihydrobacillaene, although the product of that reaction is still unknown. Our lab is using improved ultra-pressure liquid chromatography (UPLC) methodologies to detect the product of the reaction between dihydrobacillaene and the cytochrome P450 enzyme. Our most updated results will be presented in this poster.
