The primary goal of this study was the rapid purification of a recombinant Human IgG1 from a CHO cell line using Radial Flow Chromatography (RFC). The secondary goal was look at the data and how it supports Radial Flow Chromatography implementation in current intensification/integration platforms. A trend in Bioprocessing is intensification/integration with a focus on process (or step) integration and decease of operation time (intensification). Radial Flow Chromatography is an easy to implement, commercially available column exhibiting “rapid” adsorptive flow with low operational pressure that makes it readily adaptable to the intensification/integration process platforms. Radial Flow Chromatography (RFC) is a proven, fast, high recovery low pressure chromatographic technology, that has been highly effective for protein purification at commercial scale. The RFC column design makes it is extremely suitable for high throughput adsorptive separations at all process scales (lab, process pilot and manufacturing) with the added advantage it can use any commercially available resin. To support the use of Radial Flow Chromatography (RFC) for a process intensification/ integration project, a series of studies were conducted. A 100ml Sepragen Corporation Superflo® Radial Flow Column was packed with a commercially available Protein A resin. Once packed, the purification was done on a QuantaSep® 1000LX system. Studies were conducted to obtain antibody loading capacity, flow rate effect, and an exploration of overload conditions. The data and its discussion of its theoretical implementation into intensification /integration model will be presented.
