Permeability of lipid bilayers is important yet hard to investigate untill the development of surface sensitive and selective technique, second harmonic generation (SHG). Since the first application of SHG in studying the molecular dynamics on the surface of particles and liposomes by Prof. Eisenthal’s group, the transport of molecules aross lipid bilayers in vesilces, cells and bacterias has been intensively investigated. Here, we studied the permeability of lipid bilayers on the surface of vesicles ranging in size from tens of nanometers to several microns through angle resolved SHG. It was revealed that within this size range, there was notable difference in the permeability of the lipid bilayers for a dye (D289) molecule at room temperature (295K). With the temperature raised to 313 K, the cross-membrane transport of D289 was significantly promoted on all the vesicles.
