O-GlcNAc modification has been observed on over 1,000 proteins. However, the potential biochemical consequences of this posttranslaitonal modification on the vast majority of protein substrates are unknown. Discovering the effects of O-GlcNAc is complicated by a lack of tools for the installation and subsequent invesigation of site-specific O-GlcNAc marks. Here, I will disucss how my lab overcomes this limitation using synthetic protein chemsitry and how we have used our O-GlcNAc-modified proteins to make important biological discoveries.
