Proteins are the molecular machines of living systems. Their dynamics are an intrinsic part of their evolutionary selection in carrying out their biological functions. Although more difficult to observe than a static, average structure, we are steadily improving ways to observe these dynamics and form sound mechanistic connections between structure and dynamics together to explain the functions of proteins. The methods include more powerful applications in X-ray crystallography, cryo-electron microscopy, and other fields. Some examples from the author and others will be presented and reviewed as well a description of the role of the Protein Data Bank in these efforts.
