There is an urgent need for increasing LC resolution and MS compatibility for proteins because of both the increasing sophistication of biologics in the pharmaceutical industry and the realization that potential protein biomarkers for disease are low in abundance. Our group investigates the ability of thin polymer brush layers on silica to shield proteins from the silica surface to enable use of MS-compatible mobile phase additives, and also to provide versatile chemical functionalities for selectivity in separations. We find polymer brushes to be advantageous bonded phases for RPLC, HIC, WCX and HILIC of therapeutic proteins. This presentation will emphasize hydrophilic polymers for performing HILIC-MS of glycoproteins. Results include a study of the HILIC separation mechanism for a model glycoprotein, ribonuclease B, as well as applications to characterizing glycosylation of therapeutic monoclonal antibodies and improving separations of complex samples in glyco-proteomics.
