A novel water-soluble derivative of the noria macrocycle was developed through a facile, aqueous synthesis. This macrocycle contains twelve sulfonatomethyl groups on the exterior of the noria. This novel macrocycle displays high water solubility across a wide range of pH values. Distinct species containing a neutral charge, a -12 charge, and a -36 charge can be formed and isolated by increasing the pH. Binding studies were performed with various amino acids in aqueous media with the water-soluble noria and its associated rotaxane structure to study the allosteric relationship between the interior cavity and the exterior binding pockets.
