TET2 is an enzyme that catalyzes the stepwise oxidation of the common 5-methylcytosine (5mC) epigenetic mark. The H1881 residue is coordinated to the catalytic metal; the H1881R variant is implicated in several hematologic disorders. We performed molecular dynamics simulations of the H1881R variant with three different substrates, 5mC, 5hmC, and 5fC. The variant affects the overall fit of the substrate in the active site. Our results highlight the importance of H1881 for structural stability.
