We have developed a tunable, chelating, phospho-organosilane nanopolymer core, which serves as a platform for selectively targeting solid tumors via the Enhanced Permeability and Retention (EPR) effect. The polymer networks chelate ions with diagnostic and therapeutic properties, such as manganese (II) or 177lutetium (III). The polymerization conditions of the core can be tuned to give sizes from 2-40 nm, on a multi-kg scale. Suitable bio-inert coatings can be attached to optimize the pharmacokinetics for the desired application. SN132D is a 5.5 nm clinically safe manganese-based MRI-contrast agent with proven contrast enhancement of breast tumors. We are showing preliminary data from a patient enrolled in our ongoing SPAGOPIX-01 clinical study. This shows that the EPR effect can be favorably exploited in humans. SN201 is a 30 nm 177Lu-radioisotope carrying therapeutic nanoparticle, which has shown anti-tumor efficacy in mouse models of mammary gland and colon carcinoma. We also show SPECT-CT images, indicating good delivery to tumors. Synthetic routes, data on radiostability and protein-binding are also provided. The strategy to create diagnostic and therapeutic nanoparticles based on this novel polymer platform opens up a wide range of precision diagnosis and therapy applications to target several life-threatening diseases.
