Plasmid DNA yield and quality from <i>E. coli</i> fermentation can be highly dependent on the host strain and the DNA sequence. As a result, developing platform fermentation and purification processes for high DNA yield and quality can prove challenging. In addition, several months may be required to perform process development, creation and testing of a cell bank, and subsequent use for GMP manufacturing and purification of plasmid DNA. To enable rapid production of high-quality plasmid DNA, isothermal rolling circle DNA amplification (RCA) is being used to develop a scalable process for cell-free production of pDNA. Process conditions and the characterization of the RCA DNA will be discussed herein.
