Apolipoprotein E (ApoE) is a lipid transport protein that has been found to be associated with multiple diseases, including heart disease. The changes in heart health are expected to change most dramatically at surfaces which interact with the lipoproteins. In my time at Brigham Young Univeristy, we hypothesized that lipoprotein receptors and enzymes of lipid metabolism will be perturbed by changes in ApoE. In a proof-of-concept study we examined healthy mice hearts using Liquid Chromatography-Mass Spectrometry (LCMS), cryosectioning, and staining. Hearts were prepared for the LCMS using cryopulverization, and further analyzed by separating the heart tissue into the mitochondrial and cytosolic fractions. In preparation for localized sampling of affected regions we prepared slices of heart for laser capture microsampling (LCM) of the tissue followed by LC-MS/MS.A comparative study of the proteins observed using the cryopulverization technique and highlight expectations for analysis of LCM samples will be presented.
